In Multisite Settings Overrides .... is there a core or ecom we can set to when a new account is signed up for that we would need to approve before the customer can start ordering? Thank you

Assuming this is for a wholesale site, you can allow users to sign up for a basic account (which is the account type for all new accounts), and only sell to wholesale accounts. Settings can be found here:
System / Component / Settings / Account Section
Store / Component / Settings / General Store Settings

Is there a way to do a settings override to have just one multisite function this way rather than globally for all site which appears to be what these setting do?

Yes you can override the wholesale status field. On the multisite, you want to control find the Settings under the "Settings Overrides" and add core.whlactive with a value of "1" if you want it to be active on that site or "0" if you do not want the system active.

Can we have them sign up just like for a wholesale account have the application process divert over to a sign up page rather than download a pdf form?
